Overview

LY4268989, also known as MORF-057, is a **selective oral small molecule inhibitor of the integrin α4β7**. It is being developed for the treatment of **inflammatory bowel diseases (IBD)**, including **ulcerative colitis** and **Crohn’s disease**. The drug works by **blocking the interaction between α4β7 on lymphocytes and the mucosal endothelial ligand MAdCAM-1**, thus **reducing lymphocyte migration into intestinal tissues and decreasing intestinal inflammation** associated with IBD. This mechanism is clinically validated by the success of biologic inhibitors of the same pathway, such as the approved antibody therapy vedolizumab. LY4268989 is currently being evaluated in **Phase 2 clinical trials** for its efficacy and safety in patients with moderately to severely active ulcerative colitis and Crohn's disease, as well as in Phase 1 studies for pharmacokinetics and safety in healthy subjects, including Japanese and Chinese participants[1][3][4][5][7][9].
