Overview

LY4298445 is an investigational monoclonal antibody developed by Eli Lilly and Company for the treatment of autoimmune disorders, specifically systemic lupus erythematosus (SLE) and rheumatoid arthritis (RA). The drug targets the CD40 ligand (CD40L, also known as CD154), a protein primarily expressed on the surface of activated T cells. By binding to CD40L, LY4298445 prevents its interaction with the CD40 receptor on B cells and other antigen-presenting cells, thereby inhibiting the co-stimulatory signals necessary for B cell activation, antibody production, and the overall inflammatory response. As of 2024, LY4298445 is undergoing Phase 1 clinical evaluation to determine its safety, tolerability, and pharmacokinetic profile in healthy participants and patients with SLE or RA.
