Overview

LY4302814 is an investigational monoclonal antibody developed by Eli Lilly and Company, currently in Phase 1 clinical development for cardiovascular and metabolic indications, specifically dyslipidemia. The therapeutic targets Angiopoietin-like 3 (ANGPTL3), a protein primarily expressed in the liver that regulates lipid metabolism by inhibiting the enzymes lipoprotein lipase (LPL) and endothelial lipase (EL). By neutralizing ANGPTL3, LY4302814 is designed to enhance LPL and EL activity, thereby reducing circulating levels of triglycerides, low-density lipoprotein cholesterol (LDL-C), and other atherogenic lipoproteins. This mechanism is particularly relevant for patients with homozygous familial hypercholesterolemia (HoFH) or severe hypertriglyceridemia who do not achieve adequate lipid control with standard therapies.
