LY4336619 is a fully human, common light chain-based bispecific antibody targeting placental-like alkaline phosphatase 2 (ALPPL2) and CD3, constructed on an Fc-silenced IgG1 backbone. It functions as a T-cell engager, designed to recruit and activate T cells specifically at the site of ALPPL2-expressing tumor cells, thereby inducing targeted T cell-mediated cytotoxicity. ALPPL2 is a tumor-associated antigen with high expression in various solid tumors, including ovarian and endometrial cancers, but very limited expression in normal adult tissues, making it an ideal target for immunotherapy. Preclinical data indicates that LY4336619 possesses high specificity for ALPPL2 and the closely related ALPP, while avoiding other alkaline phosphatase family members such as ALPI and ALPL. It has demonstrated potent antitumor activity in xenograft models and a favorable safety profile with low cytokine release.
