Overview

LY4405094 is a small interfering RNA (siRNA) therapeutic developed by Eli Lilly for the treatment of Alzheimer's disease. It is designed to target the mRNA of the microtubule-associated protein tau (MAPT) gene, leading to its degradation and a subsequent reduction in the production of tau protein. The accumulation of hyperphosphorylated tau into neurofibrillary tangles is a core pathological feature of Alzheimer's disease and is strongly correlated with neurodegeneration and cognitive impairment. By silencing MAPT expression, LY4405094 aims to slow or halt disease progression. The drug is administered via intrathecal injection and is currently undergoing Phase 1 clinical evaluation to assess its safety, tolerability, and pharmacodynamic effects on tau levels in healthy participants and individuals with Alzheimer's disease.
