The LY6K peptide vaccine is a cancer immunotherapy targeting the tumor-associated antigen lymphocyte antigen 6 complex locus K (LY6K), which is overexpressed in various cancers including lung, esophageal squamous cell carcinoma, gastric cancer, and head-and-neck malignancies. The most studied formulation includes the HLA-A*24-restricted epitope (LY6K-177) or long peptides encompassing both cytotoxic T lymphocyte (CTL) and helper T cell epitopes. The mechanism of action involves stimulating an immune response—specifically activating CTLs and Th1-type CD4+ T cells—against tumor cells expressing LY6K. Clinical trials have demonstrated safety and immunogenicity in patients with advanced gastric cancer as well as promising results in lung and esophageal cancers[1][2]. The vaccine is typically administered with adjuvants such as Montanide ISA 51 to enhance immune activation[1]. It has been evaluated both as a single-peptide formulation (e.g., S-488210) and as part of multipeptide vaccines for broader anti-tumor activity[3][4].
