LYL119 is an investigational, next-generation autologous chimeric antigen receptor (CAR) T-cell therapy targeting ROR1, a cell surface protein highly expressed in several solid tumors. Developed by Lyell Immunopharma, LYL119 incorporates four proprietary T-cell reprogramming technologies—c-Jun overexpression, NR4A3 knockout, Epi-R manufacturing protocol, and Stim-R activation technology—to enhance CAR T-cell potency and durability by reducing exhaustion and promoting stem-like memory characteristics. Preclinical studies demonstrated that LYL119 exhibits superior cytotoxicity, sustained cytokine production upon repeated antigen stimulation, robust antitumor efficacy at low doses in xenograft models of non-small cell lung cancer (NSCLC), and improved persistence compared to earlier constructs. The product is being advanced into Phase 1 clinical trials for patients with ROR1-positive solid tumors such as platinum-resistant ovarian cancer and endometrial cancer[5][6][8][9][10].
