LYL331 is an investigational T-cell receptor (TCR) gene therapy engineered to target the cancer/testis antigen NY-ESO-1 (also known as cancer/testis antigen 1 or CTAG1B), which is frequently expressed in several solid tumors but not typically found in normal adult tissues. The therapy is designed to enhance T-cell recognition and killing of NY-ESO-1 expressing tumor cells by gene transference and replacement of endogenous T lymphocyte specificity. It was initially developed by Lyell Immunopharma, later partnered with GSK, but there was no IND filing or human dosing before development of the asset was discontinued in the preclinical stage. The program (as well as a related asset, LYL132) used Lyell’s genetic/epigenetic reprogramming technologies to try to improve the durability and vigor of TCR T cell therapy responses for solid tumor malignancies[1][3][4][5][6][7][8].
