Overview

LYL797 is an investigational, autologous chimeric antigen receptor (CAR) T-cell therapy targeting receptor tyrosine kinase-like orphan receptor 1 (ROR1). It is enhanced with genetic and epigenetic reprogramming technologies, including c-Jun overexpression and Lyell’s proprietary Epi-R manufacturing protocols, to improve T-cell proliferation, reduce exhaustion, and promote stem-like characteristics. These enhancements are designed to optimize efficacy in solid tumors by overcoming biological barriers such as T-cell exhaustion. LYL797 is being developed primarily for the treatment of ROR1-positive cancers, including triple-negative breast cancer (TNBC), non-small cell lung cancer (NSCLC), ovarian cancer, endometrial cancer, multiple myeloma, and chronic lymphocytic leukemia. The product is manufactured individually for each patient from cells obtained via leukapheresis.
