Overview

LYL845 is an investigational autologous tumor infiltrating lymphocyte (TIL) therapy developed by Lyell Immunopharma. It is enhanced using the proprietary Epi-R epigenetic reprogramming protocol, which generates populations of tumor-reactive T cells with stem-like qualities and a more favorable phenotype, including CD8+ skewing. These features are associated with improved clinical outcomes compared to conventional TIL products. The Epi-R process aims to preserve relevant T-cell clones and maintain durable stemness—enabling self-renewal, persistence, and robust anti-tumor activity. LYL845 is being evaluated in a Phase 1 clinical trial for patients with relapsed or refractory metastatic or locally advanced melanoma, non-small cell lung cancer (NSCLC), and colorectal cancer (CRC). The product has received FDA Orphan Drug Designation for the treatment of stage IIB-IV melanoma[1][2][3][6][7][9].
