Overview

Lym-1 is a murine IgG2a monoclonal antibody that specifically targets the HLA-DR10 protein, a cell surface marker present on over 80% of lymphoma cells. Developed in the early 1980s by immunizing mice with nuclei from Raji lymphoma cells, it binds to a conformational epitope on several HLA-DR subtypes with higher affinity for malignant B cells than normal B cells. The antibody has been used as an unconjugated agent and as a radioimmunoconjugate labeled with isotopes such as iodine‑131 (131I) or copper‑67 (67Cu) for radioimmunotherapy of chemotherapy-resistant non-Hodgkin lymphoma and other B-cell malignancies. Clinical trials have demonstrated tumor regression and durable remissions in patients with refractory disease. Mechanistically, when bound to its target epitope, Lym‑1 can induce apoptosis or autophagy in target cells and mediate cytotoxicity through ADCC or CDC[3][4][7][9].
