Overview

Pharis Biotec is developing a therapeutic version of the Lympho-Epithelial Kazal-Type Inhibitor (LEKTI) for the treatment of atopic dermatitis. LEKTI is a multi-domain serine protease inhibitor naturally found in the stratum granulosum of the epidermis, where it regulates the activity of kallikrein-related peptidases (KLKs) such as KLK5, KLK7, and KLK14. In atopic dermatitis, a relative deficiency or imbalance of LEKTI leads to uncontrolled protease activity, which degrades corneodesmosomes and triggers pro-inflammatory signaling through PAR2. By delivering exogenous LEKTI, Pharis Biotec aims to restore the skin's protease-antiprotease balance, repair the skin barrier, and alleviate inflammatory symptoms. The project is currently in preclinical development.
