Overview
LYN-157 is an investigational, oral, extended-release capsule containing a fixed-dose combination of memantine hydrochloride and donepezil hydrochloride. Developed by Lyndra Therapeutics (specifically Lyndra Australia Pty Ltd), it utilizes a gastric-resident dosage form designed to provide consistent, linear release of both active ingredients over a seven-day period. Memantine acts as an NMDA (N-methyl-D-aspartate) receptor antagonist, while donepezil functions as an acetylcholinesterase inhibitor. This combination targets two distinct pathological pathways in Alzheimer's disease. The long-acting formulation aims to improve patient adherence and maintain steady-state plasma concentrations compared to daily oral therapies. It has been evaluated in Phase 1 clinical trials for safety, tolerability, and pharmacokinetics in healthy volunteers.
