Overview

Lyn siRNA is a small interfering RNA designed to silence the expression of Lyn tyrosine kinase, a member of the Src family of non-receptor tyrosine kinases. In preclinical research, Lyn has been identified as a key driver of the epithelial-mesenchymal transition (EMT) and cancer metastasis. By targeting Lyn mRNA for degradation, this siRNA reduces the expression of mesenchymal markers such as vimentin, fibronectin, and Zeb-1, while concurrently increasing the expression of the epithelial marker E-cadherin. The mechanism of action involves the disruption of a Vav-Rac-Pak1 signaling cascade, which normally prevents the proteasomal degradation of SNAI proteins (Snail and Slug). Silencing Lyn leads to the degradation of these transcription factors, thereby inhibiting cell migration and invasion in various malignancies, including prostate, breast, and bladder cancers.
