Overview

Lysostaphin is a recombinant bacteriolytic enzyme (specifically a glycyl-glycine endopeptidase) originally isolated from *Staphylococcus simulans*. It specifically targets and cleaves the pentaglycine cross-bridges in the peptidoglycan layer of the *Staphylococcus aureus* cell wall, leading to rapid cell lysis and death. Lysostaphin is highly active against both dividing and non-dividing *S. aureus* cells, including methicillin-resistant *S. aureus* (MRSA) and vancomycin-resistant strains. It has been developed by several companies, including AMBI, Bionexus, Shanghai Hi-Tech Bioengineering, Ying Tai Bio-Technology, and Bharat Biotech, for the treatment of *S. aureus* infections, wound infections, and burn infections. Additionally, lysostaphin-based formulations (such as compound Staphylococcal lysostaphin mouthrinse) have been evaluated in Phase 3 clinical trials for the prophylaxis of oral mucositis in patients undergoing hematopoietic stem cell transplantation.
