Drug intelligence / Profile preview
LysSM5 is a novel bacteriophage-derived endolysin (enzybiotic) derived from the Salmonella phage vB_SenA_SM5. It is a 28.8 kDa modular protein consisting of 264 amino acids, featuring an N-terminal peptidoglycan-binding domain and a C-terminal N-acetyl-β-D-muramidase catalytic domain. LysSM5 is designed to target and degrade the peptidoglycan layer of bacterial cell walls, leading to osmotic lysis and cell death. While it exhibits potent, dose-dependent lytic and antibiofilm activity against Gram-positive bacteria like *Micrococcus luteus*, its activity against Gram-negative pathogens such as multidrug-resistant *Salmonella enterica* serovar Typhi (*S. Typhi*) is enhanced synergistically when co-administered with outer membrane permeabilizers like EDTA. LysSM5 is being developed by the Postgraduate Institute of Medical Education and Research (PGIMER), Chandigarh, India, as a potential therapeutic agent to address antimicrobial resistance.
