M0324 is a novel **bispecific antibody-based immunomodulator** designed to selectively activate the CD40 immune signaling pathway, but only in the presence of high levels of MUC1—a tumor-associated antigen commonly overexpressed in certain cancers. The drug functions as a "conditional agonist," only triggering CD40 signaling when MUC1 is present, thereby **activating dendritic cells and stimulating anti-tumor immune responses in MUC1-high tumor microenvironments**. This targeted approach aims to minimize off-tumor immune activation, boosting safety. Preclinical and early clinical data indicate strong activation of dendritic cells, increased IL-12p40 secretion, and robust elimination of tumors in MUC1-expressing cancer models, particularly in pancreatic and colorectal cancers. M0324 is currently being evaluated in phase 1 trials in advanced malignant solid neoplasms and solid tumors, as monotherapy and in combination with pembrolizumab or chemotherapy[1][2][3][4][6][9][10].
