M07-TFN is a fully human monoclonal antibody agonist targeting the human triggering receptor expressed on myeloid cells 2 (hTREM2), developed by ISAR Bioscience for the treatment of Alzheimer's disease and other neurodegenerative disorders. The antibody is uniquely engineered with a transferrin receptor (TfR) binding domain (TFN) to facilitate transcytosis across the blood-brain barrier, ensuring effective brain delivery. Upon binding to hTREM2 on microglia, M07-TFN potently activates Syk signaling and reduces TREM2 shedding, thereby enhancing microglial phagocytic activity and survival. Preclinical studies in Alzheimer's mouse models have demonstrated its ability to reduce amyloid plaque load and improve cognitive functions such as memory and spatial orientation.
