Overview

M102.4 is a fully human IgG1 monoclonal antibody developed for the treatment and post-exposure prophylaxis of infections caused by Hendra virus (HeV) and Nipah virus (NiV), which are highly lethal zoonotic henipaviruses. Discovered through a collaboration between the Uniformed Services University of the Health Sciences (USU) and the National Cancer Institute (NCI), the antibody was isolated from a large human antibody library. It specifically targets the G glycoprotein of these viruses, binding to the ephrin receptor-binding site. By occupying this site, M102.4 prevents the virus from attaching to host cell receptors (ephrin-B2 and ephrin-B3), thereby neutralizing the virus and blocking cellular entry. The antibody has been used on a compassionate use basis in over 16 individuals following high-risk exposures and has undergone Phase 1 clinical testing to evaluate its safety, tolerability, and pharmacokinetics in healthy adults.
