Drug intelligence / Profile preview
M107 is a small molecule drug candidate developed for the treatment of gastroparesis, a chronic and serious gastrointestinal disease characterized by delayed stomach emptying due to impaired stomach nerves and muscles. It is being developed by Aclipse Therapeutics. Mechanistically, M107 acts as an agonist of peroxisome proliferator-activated receptor gamma (PPARγ). By activating PPARγ, M107 lowers inflammation in the stomach and increases the population of interstitial cells of Cajal (the pacemaker cells responsible for gastric motility), thereby improving gastric emptying and reducing fibrosis at the pylorus. Uniquely among gastroparesis therapies in development or on the market, M107 aims to modify disease progression rather than only treating symptoms by switching pro-inflammatory human macrophages (M1) to anti-inflammatory macrophages (M2)[1][2].
