Overview

M127101 is a small molecule inhibitor of Signal Transducer and Activator of Transcription 3 (STAT3) developed by Maruho Co., Ltd. It was primarily investigated for the treatment of eosinophilic chronic rhinosinusitis (ECRS) with nasal polyps, a condition characterized by Type 2 inflammation and eosinophil infiltration. The drug was formulated as a 0.0003% solution for once-daily administration via a nebulizer. In a Phase 2 randomized, placebo-controlled study conducted in Japan, M127101 failed to show a significant efficacy benefit compared to placebo in reducing nasal polyps or congestion, leading to the cessation of development for this specific indication.
