m170 is a murine monoclonal IgG1 antibody that specifically binds with high affinity to aberrant carbohydrate residues on MUC-1, an epithelial mucin commonly expressed on various adenocarcinoma cells, including prostate and breast cancer. It is utilized as a radioimmunoconjugate, typically labeled with Indium-111 (111In) for imaging and Yttrium-90 (90Y) for therapeutic radiation delivery. The radioisotopes are linked to the antibody via a novel DOTA-peptide linker, which is designed to reduce radiation exposure to the liver by facilitating the clearance of the radiochelate. This targeted approach aims to deliver radiation directly to tumor sites, and it has been studied in combination with paclitaxel to enhance antitumor effects in metastatic hormone-refractory prostate cancer and advanced breast cancer. Cyclosporine has been used in trials to prevent the development of human anti-mouse antibodies (HAMA), thereby enabling the potential for multidose, fractionated therapy.
