M19-6 is a small molecule inhibitor that targets the MA3 domain of the eukaryotic translation initiation factor 4 gamma 1 (eIF4G1). By binding to this specific domain, M19-6 impairs the assembly of the eIF4F translation initiation complex, which is a rate-limiting step in mRNA translation. This inhibition leads to attenuated protein translation and suppresses the 2D and 3D growth of melanoma cells. Research indicates that M19-6 alters the expression of genes involved in apoptosis, the unfolded protein response (UPR), cell cycle regulation, and reactive oxygen species (ROS) pathways. Furthermore, it has shown synergistic effects when combined with autophagy inhibitors, leading to enhanced melanoma cell death.
