Overview

M1i is a potent small-molecule inhibitor targeting methyltransferase-like protein 1 (METTL1), an enzyme responsible for depositing tRNA 7-Methylguanosine (m7G) modification. It is being investigated as a potential therapeutic agent for acute myeloid leukemia (AML). M1i inhibits METTL1's m7G methyltransferase activity, leading to a reduction in tRNA m7G levels and disruption of the tRNA^phe_GAA/HCK/CXCR4 signaling cascade, which is crucial for leukemia stem cell (LSC) self-renewal and bone marrow homing. Preclinical studies have shown that M1i significantly reduces LSC frequency, delays leukemogenesis, and prolongs survival in various myeloid leukemia models.
