Overview

M2.9 (also known as Peak Bio-Trop2-ADC) is a preclinical antibody-drug conjugate (ADC) developed by Peak Bio for the treatment of solid tumors. It consists of a proprietary monoclonal antibody targeting Tumor-associated calcium signal transducer 2 (Trop2/TACSTD2) conjugated to PH-1, a novel splicing modulator payload derived from Thailanstatin. The ADC is designed to target Trop2, which is frequently overexpressed in epithelial-derived solid tumors. Upon internalization, the PH-1 payload exerts a dual mechanism of action: it disrupts RNA splicing, leading to mRNA decay and cancer cell death, and it induces the formation of mis-spliced protein neoepitopes that stimulate an immune response against the tumor. Preclinical data has shown single-digit nanomolar potency across multiple solid tumor indications and a favorable safety profile in primate studies.
