M24-DOX is an antibody-drug conjugate (ADC) consisting of the anti-matriptase monoclonal antibody M24 conjugated to the cytotoxic agent doxorubicin. It is designed to selectively target and treat multiple myeloma and potentially other matriptase-overexpressing malignancies. Matriptase is a type II transmembrane serine protease that is highly expressed in B-cell lymphoma, multiple myeloma, and epithelial carcinomas, but has limited distribution in normal tissues. Upon binding to matriptase on the cell surface, M24-DOX is rapidly internalized and localized within the nuclei, where doxorubicin is released to exert its cytotoxic effects by intercalating with DNA and inhibiting topoisomerase II. This targeted approach aims to increase the therapeutic index of doxorubicin while reducing systemic toxicities, particularly cardiotoxicity.
