Overview

m276 is a human monoclonal antibody that targets CD276 (also known as B7-H3), a member of the B7 family of immune checkpoint proteins. CD276 is frequently overexpressed on the surface of various solid tumors and the tumor-associated vasculature, while maintaining low expression in normal tissues, making it an attractive therapeutic target. Developed by researchers at the National Cancer Institute (NCI), m276 is particularly notable for its high-affinity binding to both human and murine versions of CD276, a property that facilitates the study of its efficacy and safety in immunocompetent mouse models. The antibody has been primarily developed as the targeting component of antibody-drug conjugates (ADCs), such as m276-MMAE (conjugated to monomethyl auristatin E) and m276-PBD (conjugated to a pyrrolobenzodiazepine dimer). These ADCs have shown potent anti-tumor activity by directly killing cancer cells and disrupting the tumor blood supply, often leading to complete tumor regression and the induction of long-term anti-tumor immunity in preclinical studies.
