Overview

M2SR is a live attenuated influenza vaccine platform developed by FluGen. It is based on a genetically engineered influenza virus in which the essential replication gene for the M2 protein has been deleted. During manufacturing, the M2 protein is supplied in trans to allow only a single round of viral replication. This design means that after intranasal administration as a spray, the virus can infect cells and express all viral proteins but cannot produce infectious progeny or cause disease. The result is robust stimulation of mucosal (local), humoral (serum antibody), and cellular (T cell) immune responses—including against conserved antigens such as hemagglutinin (HA), neuraminidase (NA), nucleoprotein (NP), and matrix protein 1 (M1). Unlike traditional inactivated or recombinant vaccines given by injection—which mainly induce serum antibodies—M2SR also elicits durable local immunity at the site of infection and cross-reactive antibodies effective against drifted strains. Clinical studies have shown an excellent safety profile with no serious adverse effects even at high doses[1][3][4][5][6].
