M2SR H3N2 is a monovalent, live-attenuated, single-replication influenza vaccine candidate developed by FluGen. It utilizes the proprietary M2SR platform, which involves the deletion of the M2 ion channel gene from the influenza virus genome. This modification allows the vaccine virus to infect host cells and express a full complement of viral antigens—inducing mucosal, systemic, and cellular immune responses—without producing new infectious progeny. This "single-replication" approach is designed to combine the safety profile of an inactivated vaccine with the robust, broad-spectrum immunity typically associated with live-attenuated vaccines. The H3N2 candidate has been evaluated in multiple Phase 1 and Phase 2 clinical trials, including human challenge studies, where it demonstrated safety and the potential for cross-protection against drifted H3N2 strains.
