**M3M4** is an investigational co-administration regimen comprising the two modified vaccinia Ankara viral-vector HIV-1 vaccines **MVA.tHIVconsv3** and **MVA.tHIVconsv4**. The components encode complementary HIVconsvX conserved-mosaic immunogens derived from conserved regions of the HIV-1 Gag and Pol proteins that differ by approximately 8% in amino-acid sequence. The regimen is intended to expand and broaden HIV-1-specific T-cell responses, particularly CD8-positive T cells directed against conserved viral regions, as part of therapeutic-vaccination strategies for people with HIV-1 receiving suppressive antiretroviral therapy. In a completed Phase 1 study, intramuscular M3M4 vaccination was well tolerated and increased the magnitude and breadth of HIVconsvX-specific T-cell responses.
