M400×M100B-vedotin is a next-generation bispecific antibody-drug conjugate (ADC) developed by GO Therapeutics. It targets tumor-specific Tn-glycoepitopes on MUC1 and MUC4, which are neoepitopes generated by altered O-glycosylation in epithelial cancers and are largely absent from healthy tissues. The ADC is engineered as a site-specific conjugate using an mc-vc-PAB-MMAE (vedotin) linker-payload system with a drug-to-antibody ratio (DAR) of 2. By simultaneously recognizing MUC1-Tn and MUC4-Tn, M400×M100B-vedotin aims to overcome tumor heterogeneity and improve selectivity in solid tumors. Preclinical studies have demonstrated potent cytotoxicity and tumor regressions in models of breast, lung, ovarian, pancreatic, and gastrointestinal cancers.
