Drug intelligence / Profile preview
M4H (formerly known as Ab6) is a humanized IgG1 monoclonal antibody that specifically targets and blocks the transient receptor potential melastatin 4 (TRPM4) channel. Developed by the National Neuroscience Institute in Singapore, M4H was generated through CDR grafting of the mouse monoclonal antibody M4M. Under hypoxic or ischemic conditions, such as those occurring during a stroke, ATP depletion leads to the abnormal activation of TRPM4, causing excessive sodium influx, cell swelling, and oncotic cell death. By blocking TRPM4, M4H prevents these pathological changes, thereby reducing blood-brain barrier leakage, cerebral edema, and neuroinflammation. Preclinical studies in humanized TRPM4 rat models of stroke reperfusion injury have demonstrated that M4H significantly reduces infarct volume and improves functional recovery.
