Drug intelligence / Profile preview
M5493 is a novel, potent, multiple-stage anthelmintic prodrug developed for the treatment and prevention of schistosomiasis. Discovered through a phenotypic screening program against schistosome parasites, M5493 is converted in vivo to its active agent to overcome bioavailability limitations. It targets a parasite transient receptor potential (TRP) ion channel of the melastatin subfamily (specifically TRPMMCLZ), which is distinct from the target of praziquantel (TRPMPZQ). Activation of this channel causes sustained calcium influx, tissue depolarization, worm paralysis, and surface damage across multiple life-cycle stages of the schistosome worm, including both immature (juvenile) and mature stages. Unlike meclonazepam, which acts on the same target, M5493 does not engage host GABAA receptors, avoiding sedating side effects.
