Drug intelligence / Profile preview
M64 (often studied as its water-soluble hydrochloride salt, M64HCl) is a novel, selective, small-molecule focal adhesion kinase (FAK) activator. Developed through structure-activity relationship (SAR) studies as a third-generation derivative of ZINC40099027, M64 directly interacts with the FAK kinase domain to stimulate FAK autophosphorylation (specifically at Tyr397) and downstream ERK1/2 signaling. This activation promotes epithelial sheet migration, cell motility, and angiogenesis. M64 is being investigated for its therapeutic potential in promoting gastrointestinal mucosal wound healing (such as in nonsteroidal anti-inflammatory drug [NSAID]-induced enteropathy, ischemic jejunal ulcers, and inflammatory bowel disease [IBD]), accelerating skin wound repair, and managing high bone mass disorders by promoting osteoclastogenesis and bone resorption.
