**M6620 + cisplatin** is a combination investigational therapy composed of berzosertib (formerly M6620, VX-970), a potent and selective inhibitor of the serine/threonine-protein kinase ATR (ataxia telangiectasia and Rad3-related), and cisplatin, a DNA-damaging chemotherapeutic agent. Berzosertib inhibits ATR, an essential regulator of the DNA damage response, thereby preventing efficient DNA repair in tumor cells. This inhibition is designed to potentiate the anticancer efficacy of cisplatin, which induces DNA crosslinking and damage. The combination is under clinical investigation primarily in advanced solid tumors, including head and neck squamous cell carcinoma and other resistant or refractory malignancies, where preclinical and early clinical evidence suggests enhanced tumor response compared to cisplatin alone.
