M6620 inhibits ATR kinase, which is important for the DNA damage response pathway, sensitizing tumor cells to DNA-damaging agents like irinotecan by disrupting DNA repair mechanisms. Irinotecan binds to topoisomerase I-DNA complexes, preventing relegation of DNA and leading to DNA damage and cell death, especially in rapidly dividing tumor cells. The combination is being investigated for treating various advanced or metastatic solid tumors, including colorectal, pancreatic, lung, and gastric cancers, with particular focus on tumors harboring TP53 mutations and defects in DNA damage response pathways[1][2][4][5].
