M7437 is an antibody-drug conjugate (ADC) developed by EMD Serono (Merck KGaA) that targets Lymphocyte antigen 6 complex, locus E (Ly6E). The drug consists of a monoclonal antibody specific for Ly6E conjugated to exatecan, a potent topoisomerase I inhibitor payload. Ly6E is a cell surface protein frequently overexpressed in various solid tumors, including non-small cell lung cancer (NSCLC), triple-negative breast cancer (TNBC), and gastric cancer, while maintaining low expression in healthy tissues. Upon binding to Ly6E on the tumor cell surface, M7437 is internalized via endocytosis, leading to the intracellular release of exatecan. This payload then inhibits DNA topoisomerase I, causing double-stranded DNA breaks and subsequent cell death. M7437 is currently being investigated in a Phase 1 clinical trial for the treatment of advanced or metastatic solid tumors with confirmed Ly6E expression.
