M7632 is an investigational antibody-drug conjugate (ADC) developed by Merck Healthcare KGaA that targets the ganglioside GD2. GD2 is a sialic acid-containing glycosphingolipid highly expressed on the surface of various tumors of neuroectodermal and epithelial origin, including neuroblastoma, melanoma, and certain sarcomas, while having limited expression in normal tissues. M7632 consists of a humanized anti-GD2 monoclonal antibody linked to a potent cytotoxic payload, specifically a topoisomerase I inhibitor (exatecan derivative), via a protease-cleavable linker. The mechanism of action involves the antibody binding to GD2 on the tumor cell surface, followed by internalization of the ADC-antigen complex and subsequent intracellular release of the payload. The released topoisomerase I inhibitor induces DNA damage and subsequent apoptosis in the target cancer cells. M7632 is currently being evaluated in Phase 1 clinical trials for patients with advanced solid tumors.
