The M9-5 aptamer is a research-stage, nuclease-resistant (2'-fluoro-modified) RNA aptamer identified as a potential diagnostic biomarker for pancreatic ductal adenocarcinoma (PDAC). Developed by researchers at Duke University, it was discovered using a "positive/negative" SELEX (Systematic Evolution of Ligands by Exponential Enrichment) strategy designed to differentiate the secretome of MiaPaCa2 pancreatic cancer cells from that of non-cancerous HPDE cells. The aptamer specifically targets cyclophilin B (PPIB), a protein found in the secretome of pancreatic cancer cells and the sera of PDAC patients. Clinical research has demonstrated that M9-5 can discriminate between the sera of PDAC patients and healthy volunteers with high sensitivity and specificity, and its binding levels appear to correlate with tumor burden, suggesting potential utility in early detection and treatment monitoring.
