M9466 + irinotecan is an investigational combination therapy being evaluated in advanced solid tumors and colorectal cancer. M9466 is an oral PARP1 inhibitor (poly[ADP-ribose] polymerase 1 inhibitor) designed to inhibit DNA repair in tumor cells, thereby enhancing the cytotoxic effect of chemotherapeutic agents. Irinotecan is a topoisomerase 1 inhibitor that interferes with DNA replication and transcription by causing DNA strand breaks. The combination is studied to exploit potential synergy through dual impairment of DNA repair pathways, potentially improving anti-tumor activity in patients for whom standard therapies have failed.
