MA-19 is a synthetic, structurally modified derivative of the soy isoflavone genistein, developed by researchers at the Johns Hopkins University School of Medicine. It was designed to enhance the antiproliferative and apoptotic effects of genistein in breast cancer cells while minimizing its alpha-estrogenic agonist activity. MA-19 acts as a selective estrogen receptor modulator (SERM), demonstrating potent growth inhibitory activity in estrogen receptor-positive (ER+) breast cancer cell lines (such as MCF-7, 21PT, and T47D) at concentrations 10- to 15-fold lower than parent genistein. Its mechanism of action involves the down-regulation of estrogen receptor alpha (ERα) expression and the up-regulation of estrogen receptor beta (ERβ), thereby increasing the ERβ/ERα ratio to suppress tumor cell proliferation and induce apoptosis.
