MA022-2037 is a half-life extended bispecific T-cell engager (BiTE) developed by Myrio Therapeutics. It is specifically designed to target solid tumors harboring the KRAS G12V mutation in patients who are HLA-A*11:01-positive. The therapeutic utilizes fully human single-chain variable fragments (scFv) that selectively recognize a 10-mer peptide derived from the KRAS G12V neoantigen when presented by the HLA-A*11:01 complex, while avoiding cross-reactivity with the corresponding wild-type KRAS peptide. By simultaneously binding the tumor-specific peptide-HLA complex and the CD3 receptor on T cells, MA022-2037 redirects T-cell cytotoxicity to eliminate KRAS G12V-expressing cancer cells. It is currently undergoing IND-enabling studies for the treatment of lung, pancreatic, and colon cancers.
