**mA20CART** is a murine chimeric antigen receptor T cell (CAR-T) product targeting integrin αvβ6, which is highly expressed in certain solid tumor types such as pancreatic ductal adenocarcinoma (PDAC). The drug is engineered using the A20 peptide derived from the foot-and-mouth disease virus (FMDV2), specific for the αvβ6 integrin. Preclinical studies have demonstrated that mA20CART cells exhibit efficient and specific on-target cytotoxicity against αvβ6-positive cells, induce interferon-gamma (IFN-γ) upon antigen stimulation, and can prolong survival in mouse models of orthotopic pancreatic cancer. As a monotherapy, it showed modest efficacy, but efficacy was enhanced in combination with oncolytic vaccinia virus (expressing IL-21) and TGF-β blockade. This approach is intended to help overcome the stromal and immunosuppressive barriers encountered in solid tumors and expand the application of CAR-T therapies beyond hematological cancers[3].
