MA3-L is an mRNA-based cancer vaccine encapsulated in lipid nanoparticles (LNPs) designed to target the α3 domain of MICA and MICB (MICA/B). Developed by ProMab Biotechnologies, the vaccine encodes the membrane-proximal α3 domain of these stress-induced ligands. Upon administration, it elicits a humoral immune response, producing antibodies that bind to the α3 domain on the surface of tumor cells. This binding sterically hinders the proteolytic cleavage and subsequent shedding of the MICA/B α1/2 domains, a common mechanism by which tumors evade immune detection. By maintaining MICA/B on the cell surface, the vaccine prevents the downregulation of the NKG2D receptor on NK cells and T cells, thereby enhancing the immune system's ability to recognize and eliminate metastatic cancer cells. Preclinical studies have demonstrated its efficacy in models of colorectal cancer, melanoma, and breast cancer.
