MA9060 (also known as SG3014) is a novel dual-action small molecule inhibitor targeting ULK3 (Unc-51 like autophagy activating kinase 3) and BRD4 (Bromodomain-containing protein 4). Developed by researchers at the Moffitt Cancer Center, MA9060 is designed to overcome chemotherapy resistance in multiple myeloma (MM). By inhibiting ULK3, the drug disrupts autophagy, a survival mechanism that refractory MM cells utilize to survive proteasome inhibitor (PI) stress. Simultaneously, its inhibition of BRD4 targets the MYC oncogene, a key driver of MM progression. Preclinical studies have demonstrated that MA9060 can reduce tumor dissemination and extend survival in animal models without overt toxicity, suggesting potential for re-sensitizing resistant disease to standard-of-care therapies.
