MAAA-9289 is a non-targeting immunoglobulin G (IgG) antibody-drug conjugate (ADC) utilized as a negative control in preclinical oncology research. It is specifically employed in studies evaluating the efficacy of therapeutic ADCs, such as patritumab deruxtecan (U3-1402), to differentiate target-specific antitumor activity from non-specific cytotoxic effects associated with the ADC's payload and linker system. By using an ADC that lacks a specific biological target in the experimental model, researchers can establish a baseline for the background toxicity and non-specific uptake of the ADC platform. In preclinical models of prostate cancer, MAAA-9289 has been used to validate the functional therapeutic targeting of HER3 by demonstrating minimal antitumor activity compared to HER3-directed agents.
