mAb 108 is a murine monoclonal antibody that specifically targets the extracellular domain of the human epidermal growth factor receptor (EGFR). It was developed as part of early research into EGFR-targeted therapies and is frequently used in preclinical studies to investigate receptor signaling, internalization, and the induction of apoptosis. In sensitive cell lines like the DiFi human colon cancer line, mAb 108 competes with natural ligands such as EGF for receptor binding, thereby inhibiting the receptor's tyrosine kinase activity and triggering the caspase-dependent apoptotic pathway (involving caspases 8, 9, and 3). While it served as a foundational tool in understanding EGFR biology, its clinical utility was largely superseded by chimeric and humanized antibodies such as cetuximab.
