Overview

mAb 14E1 is a murine monoclonal antibody that specifically targets the human epidermal growth factor receptor (EGFR). It was developed as part of a panel of anti-EGFR antibodies (including mAb 225, the precursor to cetuximab) to investigate the mechanisms of receptor inhibition and programmed cell death in cancer. mAb 14E1 binds to a distinct epitope on the extracellular domain of EGFR, competing with ligands such as EGF and transforming growth factor-alpha (TGF-α) for receptor binding. In sensitive cell lines, such as the DiFi human colon cancer line, mAb 14E1 has been shown to induce cell cycle arrest and apoptosis through the activation of the caspase cascade, specifically caspases 8, 9, and 3. While it shares the same target as clinically approved agents like cetuximab, mAb 14E1 has primarily served as a research-use-only tool and did not progress into clinical development.
