MAb 216 is a naturally occurring human IgM monoclonal antibody derived from the VH4-34 (variable heavy chain) gene. It targets a linear B-cell lactosamine antigen (specifically a poly N-acetyllactosamine epitope) on the surface of B-lymphocytes. The antibody demonstrates direct cytotoxicity against B cells through a unique mechanism involving membrane disruption and pore formation. It has been studied primarily for the treatment of relapsed or refractory B-cell acute lymphoblastic leukemia (B-ALL), both as a single agent and in combination with vincristine. Additional research has explored its potential in treating epithelial ovarian cancer (EOC).
