mAb 225 is a murine monoclonal antibody that specifically targets the extracellular domain of the human epidermal growth factor receptor (EGFR). It was developed as a potent antagonist that competes with natural ligands, such as epidermal growth factor (EGF) and transforming growth factor-alpha (TGF-α), for binding to the receptor. This competition inhibits EGFR-mediated signaling pathways, leading to cell cycle arrest in the G1 phase and, in certain sensitive cell lines like DiFi colon cancer cells, the induction of apoptosis via the activation of the caspase cascade (caspases 8, 9, and 3). mAb 225 served as the murine precursor to the chimeric antibody **cetuximab** (Erbitux), which was developed to reduce the human anti-mouse antibody (HAMA) response for clinical applications. While mAb 225 was evaluated in early clinical trials, it has largely been superseded by its chimeric and humanized descendants in therapeutic settings.
